Claymore è ambientata un mondo immaginario di tipo medievale, in cui esistono dei mostri chiamati yoma, divoratori di esseri umani e capaci di assumerne l'aspetto. Un'organizzazione altamente segreta ha creato un ordine di guerriere sovrumane per proteggere l'umanità dagli yoma; esse vengono chiamate dagli abitanti dei villaggi Claymore, dalle omonime spade che esse utilizzano. I villaggi attaccati dagli yoma assoldano le Claymore per uccidere i mostri.
